Isolation and characterization of a bacterial strain that efficiently degrades sex steroid hormones

摘要: A bacterial strain, ZY3, growing on sex steroid hormones as the sole source of carbon and energy was isolated from the sewage treatment plant of a prophylactic steroids factory. ZY3 degrades the 3-methoxy-17-hydroxy-1,3,5(10),8(9)--4-estren (MHE). This strain was preliminarily identified as sp. ZY3 according to its morphology and its 16S rRNA gene sequence. During the experimental period (72 h), the optimum temperature, pH and 3-MHE concentration for the degradation of hydride by the strain ZY3 were 35vH, 10 and 10 mg/L, respectively. The degradation rate of the sex steroid hormones increased to 87% and 85% after the addition of maltose and peptone, respectively.

摘要: In this study, the serum levels, including thyroid hormones free triiodothyronine (FT), free thyroxine(FT),thyroid stimulating hormone(TSH)among the subjects from the exposed group ( = 48) and the control group ( = 45) were detected by immuno radiometric assay (IRMA). The expression levels of TR?1, TR?1, TSHR mRNA in placentas and umbilical cords were detected by fluorescent quantitative real-time PCR (FQ-PCR). The correlations between the thyroid hormone levels in maternal serum and umbilical serum, and between the expression levels of its receptors mRNA in placentas and umbilical cords were determined. We found that the FT levels of both maternal serum and umbilical cord serum in the exposed group were lower than those in the control ( < 0.05). However, the increased TSH levels in the exposed group had statistically significance compared to those in the control group ( < 0.05). The TR?1 and TR?1 mRNA levels both in placentas and umbilical cords in the exposed group were lower than those in the control group ( < 0.05 and 0.01). However, the TSHR mRNA levels in the exposed group were significantly different compared to those in the control group ( < 0.01). The serum FT4 and TSH levels of parturient women were positively correlated with those of the newborns in both groups ( < 0.05 and 0.01). The mRNA levels of TR?1, TR?1 and TSHR in the placentas were positively correlated with those in umbilical cords in both groups ( < 0.01). The findings suggest that some environmental pollutants existing in the electronic waste (e-waste) dismantling region may affect the health of local parturient women and newborns, representing changes both in serum levels of thyroid hormones and in mRNA expression of its receptors.

Effect of biochar amendment on soil’s retention capacity for estrogenic hormones from poultry manure

摘要: Most animals, including humans, produce natural sex hormones such as estrogens: 17β-estradiol (E2) and estrone (E1). These compounds are able to disrupt the reproductive systems of living organisms at trace concentrations (ng·L ). This experiment tests the hypothesis that 1% slow pyrolysis biochar-amended sandy soil could retain significant amount of estrogens (E1, E2) from poultry manure in its second year of application. The experiment was conducted over 46 days and consisted of a series of lysimeters containing sandy soil with biochar-amended topsoil. The application rate of poultry manure was kept at 2.47 kg·m . The biochar held a significant concentration of hormone during the first year of its application. However, in the following year (current study), there was no significant retention of hormones in the biochar-amended soil. During the first year after application, the biochar was fresh, so its pores were available for hydrophobic interactions and held significant concentration of hormones. As time passed there were several biotic and abiotic changes on the surface of the biochar so that after some physical fragmentation, pores on the surface were no longer available for hydrophobic interactions. The biochar started releasing dissolved organic carbon, which facilitated greater mobility of hormones from poultry manure down the soil profile.

美洲鲥鱼卵巢发育规律和性类固醇激素变化研究

摘要:

采用组织学、形态学及电化学等技术方法,研究了人工养殖条件下美洲鲥鱼雌性亲鱼卵巢发育和性类固醇激素的周期变化规律。结果表明,美洲鲥鱼属于非同步分批产卵类型;根据卵巢外部形状大小及色泽等的不同,可将美洲鲥鱼卵巢发育过程分为6 个时期;根据卵母细胞的形态结构、卵黄物质的积累和滤泡细胞的变化,将卵子发生划分为6 个时相;卵巢周期变化过程中,肝脏指数(HSI)和肥满度(CF)变化不显著,性腺指数(GSI)与血浆中雌二醇(E2)水平表现出与性腺发育协同一致的变化规律。

Ablation of steroid receptor coactivator-3 in mice impairs adipogenesis and enhances energy expenditure

摘要: Obesity is a medical condition in which excess body fat has accumulated to an extent and may have an adverse effect on health, leading to reduced life expectancy, impaired energy homeostasis and increased health problems. The p160 steroid receptor coactivator (SRC) gene family members have been suggested to be involved in energy homeostasis, but the impact of SRC-3 ablation on white and brown adipose tissue needs to be elucidated. In the current study, we collected data and carried out morphological studies on the effect of SRC-3 deficiency on white adipose tissue (WAT) and brown adipose tissue (BAT). Primary cells were cultured to investigate the differentiation ability of both adipocytes. Western blot was applied to detect the expression of master genes governing adipogenesis and thermogenesis. We observed that SRC-3 mice were lean, with reduced WAT and decreased serum leptin levels, mainly due to the smaller white adipocyte size caused by impaired adipogenesis, presented by decreased peroxisome proliferator activated receptor (PPAR) expression. In the BAT, the lipid droplets decreased significantly in SRC-3 mice as demonstrated by histological analysis and electron microscopic observation, which could be explained by enhanced thermogenesis. The expression of thermogenic marker gene PPAR coactivator 1α and uncoupling protein-1 increased in BAT of SRC-3 mice, which proved our observations. Collectively, these results demonstrate that SRC-3 plays a key role in adipogenesis and energy expenditure.

INTERACTIONS BETWEEN ABOVE AND BELOW GROUND PLANT STRUCTURES: MECHANISMS AND ECOSYSTEM SERVICES

Interactions between above and below ground parts of plants can be considered under the (overlapping) categories of energy, material and information. Solar energy powers photosynthesis and transpiration by above ground structures, and drives most water uptake through roots and supplies energy as organic matter to below ground parts, including diazotrophic symbionts and mycorrhizas. Material transfer occurs as water and dissolved soil-derived elements transport up the xylem, and a small fraction of water moving up the xylem with dissolved organic carbon and other solutes down the phloem. The cytosolic nature of sieve tubes accounts for at least some of the cycling of K, Mg and P down the phloem. NO3 assimilation of above ground parts requires organic N transport down phloem with, in some cases, organic anions related to shoot acid-base regulation. Long-distance information transfer is related development, biotic and abiotic damage, and above and below ground resource excess and limitation. Information transfer can involve hydraulic, electrical and chemical signaling, with their varying speeds of transmission and information content. Interaction of above and below ground plant parts is an important component of the ecosystem service of storing atmospheric CO2 as organic C in soil, a process that has decreased since the origin of agriculture.

Role of water chemistry on estrone removal by nanofiltration with the presence of hydrophobic acids

摘要: Hydrophobic acid organic matter (HpoA) extracted from treated effluent has been known to improve the rejection of steroid hormone estrone by reverse osmosis (RO) and nanofiltration (NF) membranes. In this study, the effects of solution chemistry (solution pH and ionic strength) on the estrone rejection by NF membrane with the presence of HpoA were systematically investigated. Crossflow nanofiltration experiments show that the presence of HpoA significantly improved estrone rejection at all pH and ionic strength levels investigated. It is consistently shown that the “enhancement effect” of HpoA on estrone rejection at neutral and alkaline pH is attributed to the binding of estrone by HpoA macromolecules via hydrogen bonding between phenolic functional groups in feed solutions, which leads to an increase in molecular weight and appearance of negative charge. The membrane exhibited the best performance in terms of estrone rejection at pH 10.4 (compared to pH 4 and pH 7) as a result of strengthening the electrostatic repulsion between estrone and membrane with the presence of HpoA. At neutral pH level, the ability of HpoA macromolecules to promote estrone rejection became stronger with increasing ionic strength due to their more extended conformation, which created more chances for the association between estrone and HpoA. The important conclusion of this study is that increasing solution pH and salinity can greatly intensify the “enhancement effect” of HpoA. These results can be important for NF application in direct/indirect potable water reuse.

Immunoglobulin G (IgG) is the most abundant plasma glycoprotein and a prominent humoral immune mediator. Glycan composition affects the affinity of IgG to ligands and consequent immune responses. The modification of IgG N-glycosylation is considered to be one of the various mechanisms by which sex hormones modulate the immune system. Although the menstrual cycle is the central sex hormone-related physiological process in most women of reproductive age, IgG N-glycosylation dynamics during the menstrual cycle have not yet been investigated. To fill this gap, we profiled the plasma IgG N-glycans of 70 healthy premenopausal women at 12 time points during their menstrual cycles (every 7 days for 3 months) using hydrophilic interaction ultra-performance liquid chromatography (HILIC-UPLC). We observed cyclic periodic changes in the N-glycosylation of IgG in association with the menstrual cycle phase and sex hormone concentration in plasma. On the integrated cohort level, the modeled average menstrual cycle effect on the abundance of IgG N-glycosylation traits was low for each trait, with the highest being 1.1% for agalactosylated N-glycans. However, intrapersonal changes were relatively high in some cases; for example, the largest difference between theminimum and maximum values during themenstrual cycle was up to 21% for sialylated N-glycans. Across all measurements, the menstrual cycle phase could explain up to 0.72% of the variation in the abundance of a single IgG glycosylation trait of monogalactosylation. In contrast, up to 99% of the variation in the abundance of digalactosylation could be attributed to interpersonal differences in IgG N-glycosylation. In conclusion, the average extent of changes in the IgG N-glycopattern that occur during the menstrual cycle is small; thus, the IgG N-glycoprofiling of women in large sample-size studies can be performed regardless of menstrual cycle phase.
